EyePoint Pharmaceuticals' Duravyu has now failed to meet its primary endpoint in not one, but two pivotal phase 3 trials for wet age-related macular degeneration. In the LUCIA trial, patients on Duravyu gained only 2.5 letters of visual acuity versus 6.1 letters for those on aflibercept. EyePoint is now pivoting its focus to diabetic macular edema, where the drug showed more promise.
EyePoint Pharmaceuticals is facing a significant setback after its drug Duravyu (vorolanib intravitreal insert) failed to meet the primary endpoint in the phase 3 LUCIA trial — its second consecutive pivotal trial failure in wet age-related macular degeneration (AMD). The trial tested Duravyu given every 24 weeks against Eylea (aflibercept 2 mg) given every 8 weeks, with the primary goal of demonstrating noninferiority in mean change in best corrected visual acuity (BCVA) from day 1 through weeks 52 and 56.
This follows a similar failure in August 2026, when the nearly identical LUGANO trial also missed its primary endpoint. Together, the two trials enrolled more than 900 patients. Despite the primary endpoint failures, EyePoint noted that Duravyu met key secondary endpoints in both trials, including reduced treatment burden, anatomic control, and supplement-free rates.
By the Numbers:
Why it matters: Two consecutive phase 3 failures are a major blow to Duravyu's prospects in wet AMD. EyePoint is now shifting its focus to diabetic macular edema, where the drug showed positive results in the phase 2 VERONA trial — keeping some hope alive for the program.
