People are already buying unapproved versions of retatrutide online — and AI is paying attention. Researchers at the University of Pennsylvania used AI to analyze nearly 150,000 Reddit posts and found that users reported increased appetite, sleep disturbances, and elevated heart rates — strikingly different from the nausea-dominated side effects seen in clinical trials. The findings highlight how social media mining could become an early-warning system for drug safety.
Before a drug even gets FDA approval, some people are already taking it — and talking about it online. Retatrutide, an experimental next-gen GLP-1 weight-loss drug still in Phase 3 trials, is being purchased in unapproved, gray-market formulations. Researchers at the University of Pennsylvania used AI to comb through nearly 150,000 Reddit posts from ~40,000 users to document what those people are experiencing — publishing their findings in JAMA Network Open.
The results were surprising. While clinical trials flagged nausea and GI issues as the top side effects, Reddit users most commonly reported increased appetite — the opposite of what retatrutide is designed to do. Users also described sleep disturbances, elevated heart rates, and muscle-related symptoms more frequently than seen in semaglutide or tirzepatide discussions.
Key Takeaways:
Why it matters: Millions of people are obtaining experimental drugs outside regulated channels, completely invisible to standard safety monitoring. AI-driven social media analysis offers a scalable way to surface early warning signals — but rigorous follow-up research is essential before drawing clinical conclusions.
