Chemotherapy saves lives, but it often comes at a painful cost — up to 60% of patients on certain regimens develop chemotherapy-induced peripheral neuropathy (CIPN), a nerve condition causing numbness, tingling, and severe pain in the hands and feet. No preventive treatment currently exists. Now, a study published in Science suggests psilocybin — the active compound in "magic mushrooms" — could change that.
Researchers tested prophylactic psilocybin in mouse models using cisplatin, paclitaxel, and docetaxel, as well as in human-derived sensory neurons and freshly harvested nerve tissue from 29 surgical patients. Just two pre-treatment doses of psilocybin fully prevented mechanical hypersensitivity in mice, and cycle-matched dosing sustained that protection across six months of chemotherapy — without measurably impacting tumor growth.
The mechanism? Psilocybin preserved mitochondrial trafficking inside nerve axons via the 5-HT2A receptor, keeping energy supply intact in the nerve endings that chemo typically starves. Crucially, a non-hallucinogenic 5-HT2A agonist (TBG) offered comparable protection, suggesting the psychedelic effect isn't required for the therapeutic benefit.
