The team compared proximal interphalangeal (PIP) joints — frequently hit by RA — with distal interphalangeal (DIP) joints near the fingertips, which are generally spared. Using single-cell sequencing, advanced image analysis, and high-resolution 3D X-ray scanning, they found that PIP joints had more synovial tissue and a higher concentration of PI16+ fibroblasts, a specialized connective tissue cell that also responds differently to inflammatory signals.
