BARDA's Project BioShield has exercised its first procurement option under a contract with Japanese drugmaker Shionogi, ordering $36.9 million worth of cefiderocol (brand name: Fetroja) to strengthen national preparedness against drug-resistant infections and potential biological threats.
Cefiderocol is no ordinary antibiotic — it's a siderophore cephalosporin that uses a novel mechanism to penetrate the tough outer membrane of gram-negative bacteria, bypassing the resistance defenses that render many other antibiotics useless. It's FDA-approved for complicated urinary tract infections and hospital-acquired or ventilator-associated bacterial pneumonia in adults, and has become a valued option for multidrug-resistant infections.
The contract, signed in April, is worth up to $482 million and includes plans to build a US manufacturing facility, expand cefiderocol's use in children with pneumonia, and study its potential against plague (Yersinia pestis) and melioidosis (Burkholderia pseudomallei) — two naturally occurring pathogens with bioweapon potential.
